【Title】Endoplasmic Reticulum Ca?? Channels / IP? Receptors — Establishing a New Paradigm in Cellular Metabolism
【Speaker】Professor Katsuhiko Mikoshiba, Institute for Immunochemistry, ShanghaiTech University

Professor Mikoshiba discovered that the IP? receptor is the macromolecular membrane protein missing in mutant mice exhibiting ataxia, and he was the first in the world to successfully clone its cDNA. He demonstrated that IP? receptors generate intracellular calcium oscillations and proposed a novel concept that the endoplasmic reticulum—once considered merely a small intracellular organelle—regulates development, differentiation, motor function, and neural plasticity.

Through genetic analysis of patients, he identified IP? receptor type 1 as the causative gene of human spinocerebellar degeneration, revealing its deep involvement in disease. He further demonstrated that IP? receptors play key roles in cellular metabolism, including the Warburg effect in cancer, Bcl?2 family–mediated apoptosis, ER stress responses, and viral replication and infection.

In recent years, Professor Mikoshiba’s research group has succeeded in developing STAND (ultra?stable cytoplasmic antibody), a “stable intracellular antibody” that functions in soluble form without aggregation inside cells. This technology has opened new possibilities for understanding biological phenomena and treating diseases through intracellular protein manipulation. The seminar will introduce specific examples of these advances.
